      摘要 目的:探讨内质网应激相关蛋白活化转录因子6(ATF6)、CCAAT/增强子结合蛋白同源蛋白(CHOP)在针刺防治帕金森病中的作用。 方法:84只雄性SD大鼠随机分为正常组、假手术组、模型组、电针治疗7d组、电针治疗14d组、电针治疗21d组、电针治疗28d组,每组12只。模型组采取颈背部皮下注射鱼藤酮法,假手术组只注射不含鱼藤酮的等量葵花油乳化液。各电针治疗组取穴“风府”、“太冲”,每次治疗20min,每天1次,分别治疗7d、14d、21d、28d。采用免疫组化法检测大鼠黑质酪氨酸羟化酶(TH)阳性表达,运用Western blot检测大鼠黑质区ATF6、CHOP的表达。采用RT-PCR检测大鼠黑质区ATF6 mRNA、CHOP mRNA的表达。 结果:电针治疗后,各组大鼠行为学评分较模型组显著降低(P<0.01)。与正常组和假手术组相比,模型组大鼠黑质区TH的阳性表达显著降低(P<0.01),大鼠黑质区ATF6、CHOP表达显著提高(P<0.01),大鼠黑质区ATF6 mRNA、CHOP mRNA表达升高(P<0.05);与模型组相比,大鼠黑质区TH的阳性表达提高(P<0.05),ATF6、CHOP蛋白表达显著减少(P<0.01),大鼠黑质区ATF6 mRNA、CHOP mRNA表达减少(P<0.05)。 结论:电针“风府、太冲”穴能有效保护多巴胺能神经元,其机制可能与针刺治疗能抑制内质网应激途径下游的细胞凋亡因子有关。

Effects of electroacupuncture at “Fengfu” and “Taichong” on the expression of related proteins of endoplasmic reticulum stress in Parkinson's disease rats    Download Fulltext

      Abstract Objective: To explore the expression of ATF6 and CHOP related to endoplasmic reticulum stress (ERS) in rats of Parkinson's disease. Method: Eighty-four Male SD rats were randomly divided into a normal group, a sham-operation group, a model group, 4 EA groups (treatment for 7 days, 14 days, 21 days, 28 days), 12 rats in each group. PD rat model was induced by subcutaneous injection of rotenone at the nape, and rats in the sham-operation group were injected with sun-flower oil without rotenone. Rats in the EA groups were treated by EA at “Fengfu” (GV16) and “Taichong” (LR3) for 20 min, once a day. The positive cells of TH in substantia nigra of rats were detected by immunohistochemical method. The expression of ATF6, CHOP in substantia nigra were tested by Western blot. And RT-PCR method was used to evaluate the expression of ATF6 mRNA, CHOP mRNA in substantia nigra. Result: After electroacupuncture treatment, the scores of behavior in EA groups were significantly lower than those in model group (P<0.01). Comparing with the normal group and sham-operation group, the expression of ATF6, CHOP in substantia nigra area significantly increased in model group (P<0.01), the expression of ATF6 mRNA, CHOP mRNA in substantia nigra increased in model group (P<0.05); While the expression of TH in substantia nigra significantly decreased (P<0.01); Comparing with the model group, the expression of ATF6, CHOP in substantia nigra area significantly decreased in EA groups (P<0.01), the expression of ATF6 mRNA, CHOP mRNA in substantia nigra area decreased in EA groups (P<0.05), while the expression of TH in substantia nigra of rats increased (P<0.05). Conclusion: The mechanism of EA in preventing and treating Parkinson's disease may be related to that EA at “Fengfu” and “Taichong” can inhibit the apoptosis factor at the lower reaches of ERS,and then protect the dopaminergic neurons. This may be the mechanism why acupuncture may prevent and treat Parkinson's disease.
