      摘要 目的:研究虾青素对大鼠脊髓损伤后炎症反应的影响。 方法:采用改良Allen法制作脊髓损伤模型, 90只健康成年SD大鼠,随机分为虾青素组、对照组和假手术组。分光光度法检测脊髓损伤后脊髓组织的髓过氧化物酶(MPO)的活性,酶联免疫吸附测定(ELISA)法检测肿瘤坏死因子(TNF)-α、白介素(IL)-6、白介素(IL)-1β的含量,免疫组化染色检测脊髓组织MPO、TNF-α、IL-1β、IL-6的蛋白表达。干湿重法检测脊髓组织的含水量,透射电镜观察脊髓损伤后超微结构的改变并行超微结构评分,BBB评分评估脊髓损伤后的运动功能。 结果:①对照组在脊髓损伤后脊髓组织中髓过氧化物酶(MPO)的活性和TNF-α、IL-6、IL-1β的含量显著高于假手术组(P<0.05);虾青素组脊髓组织中上述指标较对照组显著降低(P<0.05),但仍显著高于假手术组(P<0.05)。②对照组脊髓组织中MPO、TNF-α、IL-6、IL-1β的蛋白表达显著高于假手术组(P<0.05);虾青素组脊髓组织中上述指标较对照组显著降低(P<0.05),但仍显著高于假手术组(P<0.05)。③对照组脊髓组织含水量显著高于假手术组(P<0.05);虾青素组脊髓组织含水量显著低于对照组(P<0.05),但仍高于假手术组。④对照组的超微结构评分显著高于假手术组(P<0.05);虾青素组的超微结构评分显著低于对照组(P<0.05),但仍高于假手术组。⑤对照组大鼠BBB评分显著低于假手术组(P<0.05),虾青素组大鼠BBB评分显著高于对照组(P<0.05),但仍显著低于假手术组。 结论:虾青素能够明显降低脊髓损伤后升高的MPO的活性和TNF-α、IL-6、IL-1β的含量,降低脊髓组织中过高的MPO、TNF-α、IL-6、IL-1β的蛋白表达,明显减轻脊髓损伤后的炎症反应,减轻脊髓水肿,明显减轻脊髓损伤后的组织病理学改变,改善了脊髓损伤大鼠的运动功能,有效地保护脊髓组织,具有明显的神经保护作用。

      Abstract Objective: To study the effect of astaxanthin on the inflammation response after spinal cord injury in rats. Method: The model of SCI is established by the modified Allen’s method. 72 healthy adult SD rats were randomly divided into the astaxanthin group, the control group and the sham operation group. Spectrophotometry for the determination of myeloperoxidase (MPO) activity of spinal cord tissue, Enzyme linked immunosorbent assay (ELISA) method for the determination of tumor necrosis factor (TNF)-α, interleukin (IL)-6,interleukin (IL)-1β, Immunohistochemical staining for detect of expression of MPO, TNF-α, IL-6 and I L-1β were used. Dry wet weight method was used for the determination of water content of spinal cord tissue. The ultrastructural changes were observed by transmission electron microscope(TEM) and the ultrastructure score were used. The motor function were evaluated by BBB score. Result: ①The myeloperoxidase (MPO) activity and the content of TNF-α, IL-6, IL-1β in spinal cord tissue in control group was significantly higher than those in astaxanthin group (P<0.05) and those in astaxanthin group was significantly higher than sham operation group (P<0.05).②The expression of MPO, TNF-α, IL-6, and IL-1β in the spinal cord in the astaxanthin group was significantly lower than that in control group (P<0.05) and significantly higher than that in sham operation group(P<0.05).③The water content of spinal cord tissue in the astaxanthin group was significantly higher than that in the sham operation group (P<0.05), and significantly lower than that in the control group (P<0.05). ④The ultrastructure score in the control group was significantly higher than that in the sham group (P<0.05), and the ultrastructure score in the astaxanthin group was significantly lower than that in the control group (P<0.05).⑤The BBB score in the astaxanthin group was significantly lower than that in the sham group (P<0.05),and significantly higher than that in the control group (P<0.05). Conclusion: astaxanthin significantly decreased the MPO activity and the content of TNF-α, IL-6, IL-1β after spinal cord injury, decrease the protein expression of MPO, TNF-α, IL-6, IL-1β in spinal cord tissue,significantly reduce the inflammatory response and spinal cord edema after spinal cord injury, significantly reduced pathological changes after spinal cord injury, improve the motor function of rats with spinal cord injury, effectively protecting the spinal cord tissue, has obvious neuroprotective effect.
